Use the mouse to connect two tiles that carry the same item. The path between them can bend at most twice, so a pair that looks close may still be unreachable.
Clearing the board is what completes a level, and the tiles that unlock a route are often the ones you would clear last. Working from the outside inward keeps the paths open, since a tile buried in the middle needs its neighbours gone before it can connect.
